实验步骤:制备47:[(3R) - 吡咯烷-3-基]氨基甲酸叔丁酯; 将制剂46的氨基甲酸酯(15.58g,46.6mmol)的乙醇(150ml)溶液在5%Pd / C(1g)存在下,在50psi,室温下氢化18小时。 加入另外的Pd / C(500mg),并将所得混合物在相同条件下氢化另外26小时。 滤除催化剂,滤液真空浓缩。 通过色谱法纯化(DCM:MeOH:0.880氨(100:0:0,体积比改为90:10:1),得到标题化合物,为浅黄色油状物(5.85g,62%)。1H NMR(400MHz,CDCl3) :δ4.56(1H,m),3.06(2H,m),2.87(1H,m),2.79(1H,m),2.78(3H,s),2.54(1H,s),1.95(1H,m ),1.73(1H,m),1.43(9H,s)ppm.MS(ESI)m / z 201 [M + H] +

实验步骤:(b)标题化合物A上面得到的化合物(14.5g,50.14mmol),Pd / C(10%,50%水溶液)(3g)和甲酸铵(12.7g,200.5mmol)在MeOH混合物(390mL)中的溶液。 将水和(45mL)加热回流5小时。 将反应物通过硅藻土过滤,滤液用AcOEt和MeOH洗涤。 将溶剂蒸发至干,得到10.6g标题化合物,为油状物(收率:100%)。1H NMR(300MHz,CDCl 3)δ:1.38(s,9H),1.72(m,1H),1.96( m,1 H),2.53(s,NH),2.80(s,3H),2.87(m,1H),2.93(m,1H),3.11(m,2H),4.58(m,1H)。
