实验步骤:将4-氧代-1,4-二氢喹啉-3-羧酸4-羟基喹啉-3-羧酸乙酯(15g,69mmol)悬浮于氢氧化钠溶液(2N,150mL)中并在回流下搅拌2小时。 冷却后,过滤混合物,用2N HCl将滤液酸化至pH4。 过滤收集所得沉淀物,用水洗涤并真空干燥,得到4-氧代-1,4-二氢喹啉-3-羧酸,为浅白色固体(10.5g,92%)。 1H NMR(DMSO-d6)δ15.34(s,1H),13.42(s,1H),8.89(s,1H),8.28(d,J = 8.0Hz,1H),7.88(m,1H),7.81( d,J = 8.4Hz,1H),7.60(m,1H)。

实验步骤:实施例2:4-氧代-1,4-二氢喹啉-3-羧酸(3)的合成:(0084)(0085)得到4-氧代-1,4-二氢喹啉-3-羧酸乙酯的悬浮液(2) )(0.5g,2.30mmol)的THF(10mL)溶液,在0℃下滴加1M LiOH(aq)水溶液(4.60mL,4.60mmol),然后将反应混合物加热至80℃。 °C 16小时。 将反应物质蒸发至干,溶于H 2 O(5mL)中,用乙醚(2×5mL)洗涤。水层用1M HCl(水溶液)酸化,过滤沉淀的化合物,真空干燥,得到所需化合物 为灰白色固体。 产量:(0.35g; 80%)1H NMR(400MHz,DMSO-d6)δ= 15.36(brs,1H),13.43(brs,1H),8.91(s,1H),8.31(d,J = 8.3Hz,1H) ),7.98-7.75(m,2H),7.62(t,J = 7.6Hz,1H)。
