实验步骤:在火焰干燥的RBF中向LAH(45.2mL,45.2mmol,1M在THF中)的悬浮液中滴加1H-吡唑-4-羧酸乙酯(3.17g,22.6mmol)在THF(20mL)中的溶液,0° C。 将反应逐渐升温至室温并在室温下搅拌过夜。 将反应混合物在冰浴中冷却,并通过依次逐滴加入1.36mL H 2 O和10mL 1MNaOH小心淬灭,然后搅拌20mm。 加入固体MgSO4,除去冰浴,在室温下继续搅拌30分钟。 通过CELITE过滤除去固体,用THF洗涤,然后用MeOH洗涤。 蒸发合并的滤液,得到(1H-吡唑-4-基)甲醇(1.75g,78.9%),为白色固体。 'HNMR(500MF-Tz,DMSO-d6)ö12.58(br.s。,1H),7.58(s,1H),7.40(s,1H),4.74(t,J = 5.5 Hz,1H),4.37( d,J = 5.2Hz,2H)。
