实验步骤:2-(4-甲酰基苯氧基)乙酸甲酯。 向4-羟基苯甲醛(3.0g,25mmol)的丙酮(61.4mL)溶液中加入K 2 CO 3(5.43g,39.3mmol),剧烈搅拌混合物。 加入甲基溴乙酸酯(2.8mL,29mmol)并将混合物在室温下搅拌3.5小时。 减压浓缩反应混合物,然后用110洗涤,用EtOAc萃取。 分离有机层,用Na 2 SO 4干燥,过滤并浓缩,得到无色油状物,其在真空下固化,分离产率为82%。 1 H NMR(400MHz,CDCl 3)3 9.87(s,111),7.82(d,J = 8.8Hz,211),6.98(d,J = 8.7Hz,211),4.70(s,211),3.79( s,3H)。 ESI-MS(m / z):195.1 [M + H]

实验步骤:实施例68(4-甲酰基苯氧基)乙酸甲酯(68)向4-羟基苯甲醛(200克,1.64摩尔),无水K 2 CO 3(600克,4.34摩尔)和碘化钠(20克,133摩尔)的混合物中加入 向无水丙酮(2500mL)中加入氯乙酸甲酯(223g,2.055mol)并回流12小时。 蒸馏丙酮并加入水(1800mL)。 将粗品68萃取到氯仿中,用Na 2 SO 4干燥,蒸馏并通过硅胶柱色谱纯化,用己烷作为洗脱剂,得到纯的68(210克,66.1%),为白色粉末。
