实验步骤:2-吡啶甲酰亚胺酸,酰肼吡啶2-氨基腙中间体向装有机械搅拌器,回流冷凝器和氮气入口/出口的1L四颈圆底烧瓶中加入2-吡啶甲腈(50g,480mmol), 水合肼(26.4g,528mmol)和乙醇(25mL)。 将所得混合物在室温下搅拌6小时,此后TLC显示一些剩余的起始原料。 加入另外7.2g(144mol)部分的水合肼,并将混合物在50℃下加热3小时。 将混合物在冰浴中冷却,过滤,然后干燥,得到40.2g固体产物。 浓缩母液,用己烷研磨并干燥,得到11.6g另外的固体产物(总共51.8g,381mmol,79%)。 1H NMR(400MHz,CDCl3)8.50(m,1H,Ar-H),8.01(d,1H,Ar-H),7.68(t,1H,Ar-H),7.28(m,1H,Ar-H) )5.38(br s,2H,-NH 2),4.62(br s,1H,-NH),2.40(br s,1H,-NH)。
