实验步骤:向1H-吲唑-6-羧酸(3.00g,18.5mmol)的N,N-二甲基甲酰胺(46mL)溶液中加入碳酸钠(2.06g,19.4mmol),然后加入碘甲烷(2.75g,1.21mL) ,19.4mmol)滴加。 将混合物在室温下搅拌过夜。 将混合物倒入半饱和碳酸氢钠中并用乙酸乙酯萃取三次。 将合并的有机层用盐水洗涤,经硫酸钠干燥,过滤并真空浓缩,得到棕色油状物。 通过快速柱色谱法(12-100%乙酸乙酯/庚烷)纯化该残余物,得到1H-吲唑-6-羧酸甲酯,为黄色固体(2.95g,90%)。 1H NMR(400MHz,CDCl3,δ):10.40(br.s,1H),8.26(s,1H),8.13(s,1H),7.84(d,J = 8.4Hz,1H),7.79(d) ,J = 8.4Hz,1H),3.96(s,3H)。
