实验步骤:化合物2a-2h,3a-3h,4a-4h,5a-5h,6a-6h,7a-7h,11a-h和12a-12h的通用方法。 将10mL二氯甲烷和搅拌棒加入100-200mg胺中。 然后加入2当量的三乙胺,并在搅拌下将反应混合物冷却至0℃。 然后,将0.9当量的氯甲酸酯,异氰酸酯或硫代异氰酸酯滴加到反应混合物中,并使其缓慢升温至室温并继续搅拌过夜。 然后将反应混合物用更多二氯甲烷稀释,用1N HCl洗涤两次,用盐水洗涤两次,用硫酸钠干燥,然后真空浓缩。 N-(2-苯乙基)氨基甲酸乙酯2d由市售材料通过常规酰化方法(氯甲酸乙酯/ TEA在DCM中)合成(方案1)。 分离化合物2d,收率为96%,无需进行色谱纯化。
