实验步骤:4-(6,7-二甲氧基 - 喹啉-4-基氧基) - 苯胺的制备添加含有甲酸钾(5.0kg),甲酸(3.0kg)和水(16.0kg)的溶液得到6,7-二甲氧基-4-(4-硝基 - 苯氧基) - 喹啉(4.0千克),10%钯碳(50%水湿,0.4千克)在四氢呋喃(TI-IF,40.0千克)中的混合物加热到大约60°C。进行添加使得反应混合物的温度保持在约60℃。当使用过程中HPLC分析(剩余起始材料少于2%,通常为15小时)确定反应完成时,过滤反应器内容物。通过在约35℃下真空蒸馏将滤液浓缩至其原始体积的一半,这导致产物沉淀。通过过滤回收产物,用水(12.0kg)洗涤,并在约50℃下真空干燥,得到标题化合物(3.0kg; 97%曲线下面积(AUC))。
