产率:86% 合成条件:at 80℃; 实验步骤:中间体7:3-(4-甲基哌嗪-1-基) - 丙-1-醇。将1-甲基哌嗪(6.99mL,63mol)溶解在甲苯(30mL)中。缓慢加入3-溴 - 丙醇(2.62mL,30mmol)并将混合物搅拌过夜。在加热至80℃并保持2小时并冷却至室温后,过滤混合物并用甲苯彻底洗涤滤饼。除去甲苯后,将残余物进行Kugelrohr蒸馏(沸点,180℃/ 2毫巴),得到无色油状物(4.08g,25.8mmol,86%)。1H NMR(CDCl3):δ= 1.70(Ψ-quint,J 5.8Hz,2H),2.26(s,3H),2.35-2.6(m,8H),2.60(Ψ-t,J = 5.8Hz),2 H),3.77(Ψ-t,J = 5.3Hz,2H),4.09(s,br。,1H)。 参考文献:[1] Patent: EP1674467, 2006, A1.
